Extinction Rebellion activists blocked off the exit ramp from the West Gate Freeway at about 8.30am.
Police have been on the scene and had reopened one lane by about 9.30am, however activists remained.
One particular person gave the impression to be locked onto a van blocking the street with a motorcycle chain.
Traffic has slowed to a crawl and Melbourne residents have been warned there may be extra to return.
Extinction Rebellion has a three-day disruption deliberate for the CBD, with individuals warned to be alert and anticipate delays.
